Colleagues:
Has anyone performed or encountered studies which suggest that Caco-2 cell
lines are somewhat "leaky", such that drug permeation is not fully
controlled by active or passive transmembrane processes?

This depends on culture conditions (e.g. days, seeding density, passage,
etc). Bit I think there are enough evidences showing that Caco-2 are not
leaky and could be used for drug transport study. In terms of active
transport or metabolic activities, caco-2 may be underexpressed for these
systems. An recent review article about Caco-2 is by Florence Delie and
Werner Rubas in Critical Rev. in Ther. Drug Delivery System 14:221-286
(1997).
