Could some one suggest me the best solublizer for nifedipine to develop
its 20mg sustained release tablet.

Some years ago I was working (analyst) with people from the galenical
department. I don't know the best solubilizer for nifedipine, but I guess
that if you ask Mrs.Isabel Guillot in E. MERCK (Mollet del Vall=E9s , Spain=
)
she will be very pleased to help you on that subject, as she was one of the
persons working with nifedipine.
I don't know the phone number, but you will easily find it in the
directory.

Nifedipine can be solubilized relatively easily in MeOH.

It all depends on the type of dosage form. If it is an oral solution for
extemporaneous use, it can be dissolved in ethanol and given if the purpose
is to prepare a solid dosage form as sustained release nifedipine should be
dissolved in chloroform, processed and the solvent should be evaporated.
Caution must be exercised because it is light sensitive. Adding an
antioxidant (metabi sulfite) and processing in dark are recommended.

That's elementary!what I need to know is how the solubilty of nifedipine
be enhanced which then can be managed through matrix to achieve desired

You can prepare a solid dispersion using PVP 40,000 or PEG 4000-1000 to
form a coprecipitate. Several papers are available that describe this
for Nifedipine (DDIP 1980;6:137-160; STP Pharma 1986;2:214-220; STP
PHarma 1986;2:299-302).
Some years ago, I used the above polymers and achieved increased
dissolution (unpublished data).
